High-Resolution Transcriptome Atlas and Improved Genome Assembly of Common Buckwheat, Fagopyrum esculentum

Common buckwheat ( Fagopyrum esculentum ) is an important non-cereal grain crop and a prospective component of functional food. Despite this, the genomic resources for this species whole family Polygonaceae, to which it belongs, are scarce. Here, we report assembly genome using long-read technology high-resolution expression atlas including 46 organs developmental stages. We found that has extremely high content transposable elements, several classes recently (0.5–1 Mya) multiplied TEs (“transposon burst”) gradually accumulated TEs. The difference in TE major factor contributing three-fold increase size F. compared with its sister tataricum . Moreover, detected differences between wild ancestral subspecies ssp. ancestrale cultivars, suggesting activity accompanied domestication. Expression profiling allowed us test hypothesis about genetic control petaloidy tepals buckwheat. showed not mediated by B-class gene activity, contrast prediction from ABC model. Based on survey profiles phylogenetic analysis, identified MYB transcription tr_18111 as potential candidate determination conical cells petaloid tepals. information patterns been integrated into publicly available database TraVA: http://travadb.org/browse/Species=Fesc/ improved transcriptomic will enable research buckwheat, practical applications.
